چکیده
Fibrin clot structure and function are major determinants of thromboembolic diseases. The study aim was to determine the impact epicatechin (a flavonoid with cardiovascular protective effects) on fibrin permeability. Plasma samples from 12 healthy subjects were incubated increasing concentrations epicatechin. Turbidity analyzed by absorbance measurement at 405 nm. nanostructure determined scanning spectrometry (wavelength 500 800 nm) fiber size electron microscopy. Permeability assess functional properties. Epicatechin addition increased maximum 0.34 ± 0.066 (vehicle) 0.35 0.077 (P = 0.1), 0.072 < 0.05) 0.065 0.5) for 1, 10 100 ?M epicatechin, respectively. radius (nm) 109.2 3.2 108.9 4.3 0.9), 110.0 3.6 109.5 3.3 0.4), distance between protofibrils 22.2 1.5 22.1 2.3 22.6 1.8 22.3 0.9) Electron microscopy confirmed these changes. permeability, expressed as Darcy's constant (Ks, cm2), 2.97 1.17 3.36 1.21 0.05), 3.81 1.41 0.01) 3.38 1.33 0.9). Polyphosphate enhances fibrin clot structure.
Polyphosphate, a linear polymer of inorganic phosphate, is present in platelet dense granules and is secreted on platelet activation. We recently reported that polyphosphate is a potent hemostatic regulator, serving to activate the contact pathway of blood clotting and accelerate factor V activation. Because polyphosphate did not alter thrombin clotting times, it appeared to exert all its proco...
متن کاملImpact of Fibrinogen Carbamylation on Fibrin Clot Formation and Stability
Carbamylation is a non-enzymatic post-translational modification induced upon exposure of free amino groups to urea-derived cyanate leading to irreversible changes of protein charge, structure and function. Levels of carbamylated proteins increase significantly in chronic kidney disease and carbamylated albumin is considered as an important biomarker indicating mortality risk. High plasma conce...
متن کاملEffects of actin filaments on fibrin clot structure and lysis.
The muscle and cytoskeletal protein actin is released from cells as a consequence of cell death and interacts with components of the hemostatic and fibrinolytic systems, including platelets, plasmin, and fibrin. We report here that incorporation of actin filaments into fibrin clots changes their viscoelastic properties by increasing their shear modulus at low deforming stresses and by nearly el...
متن کاملAβ delays fibrin clot lysis by altering fibrin structure and attenuating plasminogen binding to fibrin.
Alzheimer disease is characterized by the presence of increased levels of the β-amyloid peptide (Aβ) in the brain parenchyma and cerebral blood vessels. This accumulated Aβ can bind to fibrin(ogen) and render fibrin clots more resistant to degradation. Here, we demonstrate that Aβ(42) specifically binds to fibrin and induces a tighter fibrin network characterized by thinner fibers and increased...

Coronary artery thrombosis following plaque rupture is an important feature of myocardial infarction, and studies have highlighted the role of coagulation in this condition. Although genetic and environmental influences on the variance in coagulation protein concentrations have been reported, there are no data on the heritability of structure/function of the final phenotype of the coagulation c...
